This is a working overview of MC4 receptor, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-10-19. Anything still debated is marked as such rather than presented as settled.
Bremelanotide functions as an agonist at several melanocortin receptor subtypes, with the strongest functional activity reported at the MC4 subtype. MC4 receptors sit in hypothalamic circuits that influence appetite, energy balance, and components of sexual behaviour. Rodents lacking functional MC4 receptors show altered mating behaviour, which supports a role for this pathway in desire. The precise sequence of events connecting receptor activation to reported human effects remains only partly characterised. Because the same receptor family governs pigmentation and inflammatory signalling, selectivity is a recurring theme in pharmacological discussion.
Clinical programmes in this area have relied mainly on randomised, double-blind, placebo-controlled designs in premenopausal women. Primary endpoints usually combine a validated questionnaire covering desire domains with counts of satisfying sexual events and a separate measure of distress. Reported outcomes show statistically significant but modest average improvement over placebo, with wide individual variation. Adverse events such as nausea, flushing, and headache occur frequently and can limit tolerability. Whether short-term trial gains translate into lasting change for most users is an open question.
Lyophilised peptide is generally held below minus twenty degrees Celsius, protected from light and moisture, because hydrolysis and oxidation accumulate faster at ambient temperature. Once reconstituted, solutions are typically kept between two and eight degrees Celsius and used within a short window defined by the supplier. Repeated freeze-thaw cycles are avoided since they promote aggregation and loss of soluble material. Container material matters as well, because peptides adsorb to certain plastics and glass surfaces at low concentration. Stability figures supplied by a vendor apply only to the specific lot and buffer that were tested.
Identity and purity are usually established with reversed-phase high-performance liquid chromatography combined with mass spectrometry. A gradient of water and acetonitrile containing trifluoroacetic acid is a common mobile phase, and ultraviolet detection near 214 nanometres responds to the peptide backbone. Mass spectrometry confirms the expected molecular mass and helps reveal truncation or oxidation products. Purity is reported as a peak-area percentage, a figure that depends on the wavelength and gradient used, so values from different laboratories are not always directly comparable. Peptide mapping and amino acid analysis provide additional confirmation when required.

Compared with the related compound melanotan II, PT-141 shows markedly weaker activation of receptors tied to pigmentation. This difference stems from subtle structural variations that alter affinity distribution across receptor subtypes. Investigators propose that such selectivity produces a different side effect profile in specific applications. However, downstream consequences of prolonged receptor activation remain uncertain in the literature. Published studies do not fully agree on the duration of signaling pathway activity and the mechanisms of desensitization.
From a pharmacokinetic standpoint, the peptide is usually delivered by injection because oral bioavailability is very low; proteases in the digestive tract degrade it rapidly. After subcutaneous administration, plasma concentrations reach a peak within roughly one hour. Its elimination half-life is relatively short, with most reports placing it in the range of a few hours. Nasal formulations have also been examined, though absorption varies widely between individuals. Metabolism proceeds mainly through peptidase cleavage, and the resulting products are excreted by the kidneys.
Clinical development of bremelanotide proceeded through several reformulation attempts. An early intranasal version was discontinued, and a subcutaneous auto-injector formulation later received approval for hypoactive sexual desire disorder in premenopausal women. Approval decisions have varied by country and over time, and the product has not been universally adopted. Blood pressure elevation is a documented effect, which is why some jurisdictions require monitoring after administration. The clinical evidence base continues to evolve as additional studies are published.
Bremelanotide is a cyclic heptapeptide that acts as an agonist at melanocortin receptors. It binds MC1R, MC3R, MC4R, and MC5R, with MC4R activation considered most relevant to sexual desire pathways in the central nervous system. The molecule is a synthetic analog of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and energy regulation. Early research explored its use in tanning before attention shifted toward sexual dysfunction applications. Receptor binding affinity varies across these subtypes.
Activation of MC4R in the hypothalamus is thought to influence dopaminergic signaling, which in turn affects arousal and desire. This mechanism differs from that of phosphodiesterase type 5 inhibitors, which act primarily on vascular smooth muscle in the genital region. Because the pathway is central rather than peripheral, effects are not strictly dependent on local blood flow. The precise downstream cascade linking receptor binding to behavioral outcomes remains an area of ongoing investigation.
== Examples == Peptide signals play a role in information processing that is different from that of conventional neurotransmitters, and many appear to be particularly associated with specific behaviours. For example, in mammals oxytocin and vasopressin have striking and specific effects on social behaviours, including maternal behaviour and pair bonding. In invertebrates, CCAP has several functions including regulating heart rate, allatostatin and proctolin regulate food intake and growth, and bursicon controls tanning of the cuticle.
In 2000, the chapter on Vitamin C in the North American Dietary Reference Intake was updated to give the Recommended Dietary Allowance (RDA) as 90 milligrams per day for adult men, 75 mg/day for adult women, and setting a tolerable upper intake level (UL) for adults of 2,000 mg/day. The table here shows RDAs for the United States and Canada for children, and for pregnant and lactating women, as well as the ULs for adults. For the European Union, the EFSA set higher recommendations for adults, and also for children: 20 mg/day for ages 1–3, 30 mg/day for ages 4–6, 45 mg/day for ages 7–10, 70 mg/day for ages 11–14, 100 mg/day for males ages 15–17, 90 mg/day for females ages 15–17. For pregnancy 100 mg/day; for lactation 155 mg/day. Cigarette smokers and people exposed to secondhand smoke have lower serum vitamin C levels than nonsmokers. The reasoning is that inhalation of smoke causes oxidative damage, depleting this antioxidant vitamin. The US Institute of Medicine estimated that smokers need 35 mg more vitamin C per day than nonsmokers, but did not formally establish a higher RDA for smokers. The US National Center for Health Statistics conducts biannual National Health and Nutrition Examination Survey (NHANES) to assess the health and nutritional status of adults and children in the United States. Some results are reported as What We Eat In America. The 2013–2014 survey reported that for adults ages 20 years and older, men consumed on average 83.3 mg/d and women 75.1 mg/d. This means that half the women and more than half the men are not consuming the RDA for vitamin C.
Factor VIII is bound to VWF while inactive in circulation; factor VIII degrades rapidly when not bound to VWF. Factor VIII is released from VWF by the action of thrombin. In the absence of VWF, factor VIII has a half-life of 1–2 hours; when carried by intact VWF, factor VIII has a half-life of 8–12 hours. VWF binds to collagen, e.g., when collagen is exposed beneath endothelial cells due to damage occurring to the blood vessel. Endothelium also releases VWF which forms additional links between the platelets' glycoprotein Ib/IX/V and the collagen fibrils VWF binds to platelet GpIb when it forms a complex with gpIX and gpV; this binding occurs under all circumstances, but is most efficient under high shear stress (i.e., rapid blood flow in narrow blood vessels, see below). VWF binds to other platelet receptors when they are activated, e.g., by thrombin (i.e., when coagulation has been stimulated). VWF plays a major role in blood coagulation. Therefore, VWF deficiency or dysfunction (von Willebrand disease) leads to a bleeding tendency, which is most apparent in tissues having high blood flow shear in narrow vessels. From studies it appears that VWF uncoils under these circumstances, decelerating passing platelets. Recent research also suggests that von Willebrand Factor is involved in the formation of blood vessels themselves, which would explain why some people with von Willebrand disease develop vascular malformations (predominantly in the digestive tract) that can bleed excessively.

An IDF spokesperson stated the attack was "an official declaration of war by Hezbollah" and vowed to "neutralize" the threat. Israel struck southern Lebanon, Beirut, and the Beqaa Valley, with the IDF saying it had killed the head of Hezbollah's intelligence Hussein Makled and Adham Adnan al-Othman, the head of Palestinian Islamic Jihad's armed wing, the Al-Quds Brigades. The Royal Air Force station at Akrotiri, Cyprus, was targeted by a drone strike causing damage to a runway. Another strike on Cyprus led to Greece announcing that it would deploy frigates and F-16s to defend Cyprus from strikes by Iran. The pro-Iran militant group Iraqi Guardians of Blood Brigades claimed responsibility for attacks on the US Victory Base near Baghdad International Airport and Erbil in the Kurdistan Region of Iraq. The Islamic Resistance in Iraq claimed responsibility for 23 drone strikes on US assets in Erbil.
Top-grain leather includes the outer layer of the hide, known as the grain, which features finer, more densely packed fibers, resulting in strength and durability. Depending on thickness, it may also contain some of the more fibrous under layer, known as the corium. Types of top-grain leather include: Full-grain leather contains the entire grain layer, without any removal of the surface. Rather than wearing out, it develops a patina during its useful lifetime. It is usually considered the highest quality leather. Furniture and footwear are often made from full-grain leather. Full-grain leather is typically finished with a soluble aniline dye. Russia leather is a form of full-grain leather. Corrected grain leather has the surface subjected to finishing treatments to create a more uniform appearance. This usually involves buffing or sanding away flaws in the grain, then dyeing and embossing the surface. Nubuck is top-grain leather that has been sanded or buffed on the grain side to give a slight nap of short protein fibers, producing a velvet-like surface. Split leather is created from the corium left once the top-grain has been separated from the hide, known as the drop split. In thicker hides, the drop split can be further split into a middle split and a flesh split. Bicast leather is split leather that is coated with a layer of polyurethane or vinyl with an embossed texture. This gives it the appearance of a grain. It is slightly stiffer than top-grain leather but has a more consistent texture.
Mice resulting from ordinary breeding and inbreeding: Non-obese diabetic (NOD) mice, which develop diabetes mellitus type 1. Murphy Roths large (MRL) mice, with unusual regenerative capacities Japanese waltzing mice, which walk in a circular pattern due to a mutation adversely affecting their inner ears Immunodeficient nude mice, lacking hair and a thymus: these mice do not produce T lymphocytes; therefore, they do not mount cellular immune responses. They are used for research in immunology and transplantation. Severe combined immunodeficiency (SCID) mice, with an almost completely defective immune system FVB mice, whose large litter sizes and large oocyte pronuclei expedite use in genetic research Toxic milk mice, which fail to recruit nutrient copper into milk causing pup death. It is caused by an autosomal recessive mutation tx which arose in an inbred. Theophilos et al. 1996 found this to be genetic and localized to chromosome 8, near the centromere.
=== Obesity and diabetes === TGF-β/SMAD3 signaling pathway is important in regulating glucose and energy homeostasis and might play a role in diabetic nephropathy. As noted above in the section about macrophages, loss of TGF-β signaling in obesity is one contributor to the inflammatory milieu generated in the case of obesity.

==== Non-volatile residue ==== Another type of contamination in UPW is dissolved inorganic material, primarily silica. Any dissolved inorganic material has the potential to remain on the wafer as the UPW dries. Nonvolatile material can be detected by using nebulizer to create small droplets liquid suspended in a stream of gas. These droplets are dried an aerosol of non-volatile residue particles that can be counted by light scattering techniques.
Alpha 2-antiplasmin (or α2-antiplasmin or plasmin inhibitor) is a serine protease inhibitor (serpin) responsible for inactivating plasmin. Plasmin is an important enzyme that participates in fibrinolysis and degradation of various other proteins. This protein is encoded by the SERPINF2 gene.
α'-Methyletonitazene is a benzimidazole derivative which is an opioid designer drug. It was the most potent compound identified in a series of etonitazene analogues tested for structure-activity relationship studies on this emerging class of designer drugs, though was still slightly less potent than etonitazene itself.
=== Pharmacokinetics === Loxapine is metabolized to amoxapine, as well as its 8-hydroxy metabolite (8-hydroxyloxapine). Amoxapine is further metabolized to its 8-hydroxy metabolite (8-hydroxyamoxapine), which is also found in the blood of people taking loxapine. At steady-state after taking loxapine by mouth, the relative amounts of loxapine and its metabolites in the blood is as follows: 8-hydroxyloxapine > 8-hydroxyamoxapine > loxapine. The pharmacokinetics of loxapine change depending on how it is given. Intramuscular injections of loxapine lead to higher blood levels and area under the curve of loxapine than when it is taken by mouth.
In November 1944, the division and several attached units were redesignated as Panzerkorps Großdeutschland. By March 1945, the Panzer Grenadier Division Großdeutschland had been reduced to around 4,000 men after the Battle of Memel. It had only one Sturmgeschütz III self-propelled gun, one Panzer IV, five Panthers, and six Tiger Is by 15 March 1945. By 25 April, the division was engaged in heavy fighting in the battles around Pillau. Eight hundred men of the division were evacuated on ferries via the Baltic Sea and surrendered to British forces in Schleswig-Holstein on 9 May. The rest were either killed or captured during the fighting in Pillau or surrendered to Soviet forces on 9 May on the Vistula spit.

The approved formulation is given by subcutaneous injection and is used on an as-needed basis rather than on a fixed daily schedule. An intranasal version was studied earlier but did not reach the same stage of development. Route of delivery strongly affects how quickly the peptide appears in circulation.
It means the product is taken shortly before anticipated activity instead of every day. This pattern reduces total exposure and shapes how safety monitoring is planned. It also means effects are expected to be temporary rather than cumulative.
Desire is assessed through validated self-report questionnaires, sometimes paired with event counts and a distress scale. These instruments capture subjective experience, so results depend on how participants interpret the questions. No laboratory marker currently substitutes for such reporting.
Purity is normally expressed as a percentage of total peak area from a chromatographic run. The value depends on the column, gradient and detection wavelength chosen. Results generated under different conditions are therefore not always interchangeable.
