A practical reference on bremelanotide: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.
Reviewed 2026-02-13. Anything still debated is marked as such rather than presented as settled.
Bremelanotide is a synthetic cyclic heptapeptide that acts on a family of G-protein-coupled receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide associated with pigmentation and several central signalling pathways. A lactam bridge constrains the ring and slows enzymatic breakdown, which distinguishes it from the linear parent molecule. Research interest moved over time from pigment biology toward central nervous system effects, particularly circuits connected to sexual desire. Parenteral delivery is used because oral bioavailability is poor.
Early clinical work used an intranasal formulation, and later programmes switched to subcutaneous delivery for more consistent absorption. A subcutaneous product received regulatory approval in the United States in 2019 for premenopausal women with acquired, generalised hypoactive sexual desire disorder. Approval followed phase 3 trials in which active treatment separated from placebo on desire and distress measures, though the average difference was modest. Labeling carries a caution about transient blood pressure elevation, so cardiovascular history is assessed before prescribing. Questions about durability of benefit beyond several months remain open.
Published discussion sits at the intersection of peptide chemistry, neuroendocrinology and sexual medicine. Trial reports emphasise change scores on validated instruments, while mechanistic papers focus on hypothalamic circuits and receptor selectivity. Because placebo response in this field is large, effect sizes are usually reported with confidence intervals rather than as isolated averages. Reviews note that female and male data sets are not interchangeable and should be read separately. Diagnostic terminology has been revised over time, which complicates comparison between older and newer studies.
The melanocortin system comprises five G protein-coupled receptors, designated MC1 through MC5, that signal mainly through cyclic AMP accumulation. MC1R and MC2R are associated with pigmentation and adrenal steroid production, while MC3R and MC4R are expressed in the central nervous system and influence energy balance and behavior. MC5R appears in exocrine tissues. Natural agonists include alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone, and endogenous antagonists such as agouti-related protein modulate the same sites. This receptor family provides the framework within which bremelanotide activity is described.
Bremelanotide acts as an agonist at several melanocortin receptors, with the strongest reported activity at MC4R and measurable activity at MC1R and MC3R. Because MC4R is expressed in hypothalamic and limbic circuits, the proposed mechanism links receptor activation to modulation of central pathways involved in desire rather than to direct effects on peripheral genital tissue. The precise downstream steps remain incompletely characterized, and evidence for the involvement of specific neurotransmitters is suggestive rather than settled. Nausea and blood pressure elevation reported during trials are consistent with melanocortin signaling outside the intended target circuit.
Compared with melanotan II, bremelanotide is a smaller cyclic peptide with a more constrained backbone, which affects receptor selectivity and metabolic stability. Published descriptions give a plasma half-life on the order of a few hours after subcutaneous administration, with elimination through hepatic and renal routes and limited plasma protein binding. Central access is inferred from effects observed in animal models, although direct measurement in humans is limited. Handling and storage requirements follow from the peptide backbone, which is susceptible to hydrolysis and oxidation.

Bremelanotide, developed under the code PT-141, is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone. Its structure is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, with a lactam bridge joining the aspartate and lysine side chains. The molecule has the formula C50H68N14O10 and a monoisotopic mass near 1025 daltons. It is commonly prepared as the acetate salt and appears as a white to off-white lyophilised powder in solid form. The free acid is the pharmacologically relevant species, while the counter-ion improves handling and dissolution.
Early research on PT-141 grew out of work on melanotan II, a related cyclic peptide studied for pigmentation. Investigators observed that centrally acting melanocortin agonists also influenced sexual behaviour in animal models, and the programme shifted toward that endpoint. A nasal formulation was evaluated in clinical trials but showed inconsistent absorption, and later studies used subcutaneous administration instead. Regulatory approval in the United States followed in 2019 for a defined population of premenopausal women with acquired, generalised hypoactive sexual desire disorder. That approval was specific to that group rather than a broad indication.

===== Personal training to increase happiness ===== The easiest and best possible way to increase one's happiness is by doing something that increases the ratio of positive to negative emotions. Contrary to some beliefs, in many scenarios, people are actually very good at determining what will increase their positive emotions. There have been many techniques developed to help increase one's happiness. A first technique is known as the "Sustainable Happiness Model (SHM)." This model proposes that long-term happiness is determined upon: (1) one's genetically determined set-point, (2) circumstantial factors, and (3) intentional activities. Lyubomirsky, Sheldon and Schkade suggest to make these changes in the correct way in order to have long-term happiness. Another suggestion of how to increase one's happiness is through a procedure called "Hope Training." Hope Training is primarily focused on hope due to the belief that hope drives the positive emotions of well-being. This training is based on the hope theory, which states that well-being can increase once people have developed goals and believe themselves to achieve those goals. One of the main purposes of hope training is to eliminate individuals from false hope syndrome. False hope syndrome particularly occurs when one believes that changing their behavior is easy and the outcomes of the change will be evidenced in a short period of time.
== Career == After completing his postdoctoral fellowship in France, in 1969, Watson returned the United States and held a position as an Assistant Professor in the Department of Pharmacology at Vanderbilt University, in Nashville Tennessee. Jack was promoted to Associate Professor with tenure in 1974. While at Vanderbilt, Watson published the first edition of Introduction to Mass Spectrometry: Biomedical, Environmental, and Forensic Applications in 1976. It was the first book to include journal titles as part of the cited literature. Harold G. (Harry) Walsh had just joined the ACS as director of the Short Course program. Walsh approached Watson and asked him to teach a course. Walsh also asked that Watson select someone from the mass spectrometry industry to co-teach the course. Watson had met O. David Sparkman, an American working for the French Gas Chromatography/Mass Spectrometry company, Riber, in Paris, a few months earlier. Watson asked Sparkman to contribute to the data systems part of the course. They taught the first session at the annual Pittsburgh Conference on Analytical Chemistry and Applied Spectroscopy in the Spring of 1978. They taught the course two more times that year at the annual ACS meetings and continued teaching into the first decade of the next millennium. In 1980 Watson accepted a joint appointment in the Departments of Biochemistry and Chemistry at Michigan State University, East Lansing Michigan. He also became the director (Principal Investigator) of the National Institutes of Health (NIH) P41 Regional Resource in Mass Spectrometry at MSU.

Celloscope automated cell counter was developed in the 1950s for enumeration of erythrocytes, leukocytes, and thrombocytes in blood samples. Together with the Coulter counter, the Celloscope analyzer can be considered one of the predecessors of today's automated hematology analyzers, as the principle of the electrical impedance method is still utilized in cell counters installed in clinical laboratories around the world.
(R(H)N)2CS + HgO → (RN)2C + HgS + H2O This reaction can often be conducted as stated, even though carbodiimides react with water. In some cases, a dehydrating agent is added to the reaction mixture. The dehydration of N,N'-dialkylureas gives carbodiimides:

==== Sabellid polychaetes ==== The sabellid polychaetes, or feather duster worms, have cartilage tissue with cellular and matrix specialization supporting their tentacles. They present two distinct extracellular matrix regions. These regions are an acellular fibrous region with a high collagen content, called cartilage-like matrix, and collagen lacking a highly cellularized core, called osteoid-like matrix. The cartilage-like matrix surrounds the osteoid-like matrix. The amount of the acellular fibrous region is variable. The model organisms used in the study of cartilage in sabellid polychaetes are Potamilla species and Myxicola infundibulum.
=== Pharmacokinetics === In healthy individuals, the plasma clearance of methocarbamol ranges between 0.20 and 0.80 L/h/kg. The mean plasma elimination half-life ranges between 1 and 2 hours, and the plasma protein binding ranges between 46% and 50%. The elimination half-life was longer in the elderly, those with kidney problems, and those with liver problems.

Liu Wenhui (simplified Chinese: 刘文辉; traditional Chinese: 劉文輝; pinyin: Liú Wénhuī; Wade–Giles: Liu Wen-hui; 10 January 1895 – 24 June 1976) was a Chinese general and warlord of Sichuan and Xikang provinces (Sichuan clique). At the beginning of his career, he was aligned with the Kuomintang (KMT), commanding the Sichuan-Xikang Defence Force from 1927 to 1929. In 1929, he became Sichuan governor before being ousted by a coalition of warlords in 1933; from there, he controlled the western part of Sichuan province, then known as Xikang, ruling the area as a personal fiefdom. He maintained his autonomy from the Nationalist government and pursued development programs in the province, which he funded through the trade of opium and by maintaining cordial relations with Chiang Kai-shek during the Second Sino-Japanese War. In 1949 he defected to the Communist forces of Mao Zedong, and went on to hold high office in the new People's Republic of China, serving as Minister of Forestry (1959–1967), member of the National People's Congress, member of the National Committee of the Chinese People's Political Consultative Conference, and member of the Central Committee of the Revolutionary Committee of the Chinese Kuomintang.

It is a synthetic cyclic peptide that activates melanocortin receptors. It is given by injection and was approved in the United States in 2019 for a specific low-desire diagnosis in premenopausal women. It is not a hormonal therapy.
Earlier approaches were largely hormonal or psychological in focus, whereas this compound acts on central melanocortin signalling. It is not a vasodilator and does not share the mechanism of phosphodiesterase inhibitors. Head-to-head comparative data are limited.
Placebo responses in desire trials are large, so the average drug-placebo separation is small. Reporting therefore relies on validated questionnaires with statistical ranges. Individual responses vary widely.
Reported activity is highest at MC4R, with lower potency at MC1R and MC3R. The MC4R interaction is generally treated as the most relevant to its central effects. Selectivity is not absolute, and activity across the family is dose-dependent.
