Compared with melanotan II, bremelanotide is a smaller cyclic peptide with a more constrained backbone, which affects receptor selectivity and metabolic stability. Published descriptions give a plasma half-life on the order of a few hours after subcutaneous administration, with elimination through hepatic and renal routes and limited plasma protein binding. Central access is inferred from effects observed in animal models, although direct measurement in humans is limited. Handling and storage requirements follow from the peptide backbone, which is susceptible to hydrolysis and oxidation.
The melanocortin system comprises five G protein-coupled receptors, designated MC1 through MC5, that signal mainly through cyclic AMP accumulation. MC1R and MC2R are associated with pigmentation and adrenal steroid production, while MC3R and MC4R are expressed in the central nervous system and influence energy balance and behavior. MC5R appears in exocrine tissues. Natural agonists include alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone, and endogenous antagonists such as agouti-related protein modulate the same sites. This receptor family provides the framework within which bremelanotide activity is described.
Bremelanotide acts as an agonist at several melanocortin receptors, with the strongest reported activity at MC4R and measurable activity at MC1R and MC3R. Because MC4R is expressed in hypothalamic and limbic circuits, the proposed mechanism links receptor activation to modulation of central pathways involved in desire rather than to direct effects on peripheral genital tissue. The precise downstream steps remain incompletely characterized, and evidence for the involvement of specific neurotransmitters is suggestive rather than settled. Nausea and blood pressure elevation reported during trials are consistent with melanocortin signaling outside the intended target circuit.
Receptor activation in hypothalamic and limbic circuits is the mechanism most often cited for the observed effects on sexual desire. Signalling through MC4R couples to Gs proteins and raises intracellular cyclic AMP, which in turn modulates dopaminergic tone in reward-related pathways. Because the peptide reaches the central nervous system after subcutaneous administration, peripheral vascular changes are regarded as secondary rather than primary. The precise neural circuits that translate receptor occupancy into behavioural change remain incompletely mapped, and published accounts describe the pathway in general terms rather than as a fully resolved sequence.
Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.
PT-141 is the research code for bremelanotide, a cyclic heptapeptide derived from alpha-melanocyte-stimulating hormone. The molecule belongs to the melanocortin receptor agonist family and shows highest affinity for the MC4 receptor subtype, with weaker activity at MC1, MC3 and MC5. Its structure retains the core His-Phe-Arg-Trp sequence that defines melanocortin recognition, while cyclization and terminal modifications improve metabolic stability relative to the parent hormone. Early work classified the compound as a centrally acting agent rather than a peripherally acting vasodilator, which shaped subsequent development priorities.
| Property | Value | Notes |
|---|---|---|
| Primary receptor target | MC4R | Highest reported agonist potency within the family |
| Secondary receptor activity | MC1R and MC3R | Lower potency than at MC4R |
| Elimination half-life | About 2 to 3 hours | Measured after subcutaneous administration |
| Plasma protein binding | Approximately 44 percent | Species- and assay-dependent |
| Route of administration | Subcutaneous injection | Reviewed product uses a single-use device |
From a pharmacokinetic standpoint, the peptide is usually delivered by injection because oral bioavailability is very low; proteases in the digestive tract degrade it rapidly. After subcutaneous administration, plasma concentrations reach a peak within roughly one hour. Its elimination half-life is relatively short, with most reports placing it in the range of a few hours. Nasal formulations have also been examined, though absorption varies widely between individuals. Metabolism proceeds mainly through peptidase cleavage, and the resulting products are excreted by the kidneys.
PT-141 initiates cellular signaling by binding to specific subtypes within the melanocortin receptor family. These receptors belong to the G protein-coupled receptor superfamily, and activation raises intracellular cyclic adenosine monophosphate levels. This cascade ultimately influences neuronal circuits in the central nervous system that are associated with sexual desire and arousal. Research indicates the compound's action concentrates in hypothalamic regions rather than peripheral tissues, which helps explain some observed pharmacological features. The selectivity of receptor binding underlies its functional differences.
Reverse-phase high-performance liquid chromatography is the standard method for assessing purity. Mass spectrometry confirms molecular identity and detects sequence variants or truncation products. Ultraviolet absorbance at 214 or 280 nm is used for quantification, with the choice depending on the peptide sequence. Method validation typically addresses linearity, limit of detection, and precision across a defined concentration range. Impurity profiling may also employ ion-exchange or size-exclusion chromatography, and these techniques complement one another.
Certificate of analysis documents usually report purity percentage, molecular weight, and appearance. Researchers verify identity by comparing observed and theoretical mass values. Chain-of-custody records and batch numbers help trace material from source to experiment. Independent verification of supplier claims is considered good practice when material identity is critical to a study. Records should include the storage history of each aliquot, and unexpected deviations in appearance or solubility warrant re-testing before use.
PT-141 is the research designation for bremelanotide, a synthetic cyclic heptapeptide developed as a melanocortin receptor agonist. The peptide contains seven amino acids with an internal lactam bridge that constrains the backbone into a stable ring. Cyclisation of this kind improves resistance to enzymatic degradation relative to linear analogues. The compound was first explored for effects on melanin production, because several melanocortin receptors influence pigmentation. Its later association with sexual desire emerged from observations made during that early work, and the molecule became the subject of a separate development programme.
Bremelanotide is a moderately large peptide with a molecular mass near 1025 daltons. In lyophilised form it appears as a white to off-white powder and is freely soluble in water and other polar solvents. The intact lactam ring is essential for receptor affinity, while linearised fragments bind far more weakly. Solutions are sensitive to extremes of pH and to prolonged exposure to light and heat, so handling typically involves buffered conditions and cold storage. Its short plasma half-life reflects rapid distribution and clearance rather than chemical breakdown inside the vial.
Lyophilised peptide is generally held below minus twenty degrees Celsius, protected from light and moisture, because hydrolysis and oxidation accumulate faster at ambient temperature. Once reconstituted, solutions are typically kept between two and eight degrees Celsius and used within a short window defined by the supplier. Repeated freeze-thaw cycles are avoided since they promote aggregation and loss of soluble material. Container material matters as well, because peptides adsorb to certain plastics and glass surfaces at low concentration. Stability figures supplied by a vendor apply only to the specific lot and buffer that were tested.
Identity and purity are usually established with reversed-phase high-performance liquid chromatography combined with mass spectrometry. A gradient of water and acetonitrile containing trifluoroacetic acid is a common mobile phase, and ultraviolet detection near 214 nanometres responds to the peptide backbone. Mass spectrometry confirms the expected molecular mass and helps reveal truncation or oxidation products. Purity is reported as a peak-area percentage, a figure that depends on the wavelength and gradient used, so values from different laboratories are not always directly comparable. Peptide mapping and amino acid analysis provide additional confirmation when required.
Regulatory status varies by jurisdiction, where approved prescription products, compounded preparations and research-grade material are treated as distinct categories with different documentation requirements. Suppliers of research material commonly issue a certificate of analysis listing purity, identity and sometimes endotoxin content. Independent verification by a third-party laboratory is often recommended because self-reported figures are difficult to check. Literature discussions usually state the source, purity and storage conditions of the material used, since these details affect reproducibility. Analysts note that a reported purity figure does not by itself describe biological activity.
=== Indigenous land claims === Belize backed the United Nations (UN) Declaration on the Rights of Indigenous Peoples in 2007, which established legal land rights to indigenous groups. Other court cases have affirmed these rights like the Supreme Court of Belize's 2013 decision to uphold its ruling in 2010 that acknowledges customary land titles as communal land for indigenous peoples. Another such case is the Caribbean Court of Justice's (CCJ) 2015 order on the Belizean government, which stipulated that the country develop a land registry to classify and exercise traditional governance over Mayan lands. Despite these rulings, Belize has made little progress to support the land rights of indigenous communities; for instance, in the two years after the CCJ's decision, Belize's government failed to launch the Mayan land registry, prompting the group to take action into its own hands. The exact ramifications of these cases need to be examined. As of 2017, Belize still struggles to recognize indigenous populations and their respective rights. According to the 50-page voluntary national report Belize created on its progress toward the UN's 2030 Sustainable Development Goals, indigenous groups are not factored into the country's indicators whatsoever. Belize's Maya population is only mentioned once in the entirety of the report.
There are more than 50 types of treatments for CTS with varied levels of evidence and recommendations across healthcare guidelines, with evidence most strongly supporting surgery, steroids, splinting for wrist positioning, and physical or occupational therapy interventions. When selecting treatment, it is important to consider the severity and chronicity of the CTS pathophysiology and to distinguish treatments that can alter the natural history of the pathophysiology (disease-modifying treatments) and treatments that only alleviate symptoms (palliative treatments). The strongest evidence for disease-modifying treatment in chronic or severe CTS cases is carpal tunnel surgery to change the shape of the carpal tunnel. The American Academy of Orthopedic Surgeons recommends proceeding conservatively with a course of nonsurgical therapies tried before release surgery is considered. A different treatment should be tried if the current treatment fails to resolve the symptoms within 2 to 7 weeks. Early surgery with carpal tunnel release is indicated where there is evidence of median nerve denervation or a person elects to proceed directly to surgical treatment. Recommendations may differ when carpal tunnel syndrome is found in association with the following conditions: diabetes mellitus, coexistent cervical radiculopathy, hypothyroidism, polyneuropathy, pregnancy, rheumatoid arthritis, and carpal tunnel syndrome in the workplace. CTS related to another pathophysiology is addressed by treating that pathology.
== On-line == On-line mass spectrometry was develop to solve some of the limitations and problem that develop from off-line analysis, such as evaporation and chemical reactions of particles in the filters during long analysis time. On-line Mass spectrometry solves these problems through the collection and analysis of aerosol particles in real time. On-line instruments are very portable and allow for spatial variability to be examined. These portable instruments can be put on many different platforms such as boats, planes, and mobile platforms (e.g. car trailers). An example of this is in the picture at the beginning with the instrumentation attached to an aircraft. Like off-line, on-line mass spectrometry has many different type of instruments, which can be broken up into two types; instruments that measures the chemistry of the particle ensemble (bulk measurement) and those that measure the chemistry of individual particles (single-particle measurement). Thus based on analytical need different instrumentation is used in analysis of the aerosol particles.
Refrigeration, continually operated, typically consumes up to 50% of the energy used by a supermarket. Doors, made of glass to allow inspection of contents, improve efficiency significantly over open display cases, which use 1.3 times the energy.

=== Catabolic pathway (catabolism) === A catabolic pathway is a series of reactions that bring about a net release of energy in the form of a high energy phosphate bond formed with the energy carriers adenosine diphosphate (ADP) and guanosine diphosphate (GDP) to produce adenosine triphosphate (ATP) and guanosine triphosphate (GTP), respectively. The net reaction is, therefore, thermodynamically favorable, for it results in a lower free energy for the final products. A catabolic pathway is an exergonic system that produces chemical energy in the form of ATP, GTP, NADH, NADPH, FADH2, etc. from energy containing sources such as carbohydrates, fats, and proteins. The end products are often carbon dioxide, water, and ammonia. Coupled with an endergonic reaction of anabolism, the cell can synthesize new macromolecules using the original precursors of the anabolic pathway. An example of a coupled reaction is the phosphorylation of fructose-6-phosphate to form the intermediate fructose-1,6-bisphosphate by the enzyme phosphofructokinase accompanied by the hydrolysis of ATP in the pathway of glycolysis. The resulting chemical reaction within the metabolic pathway is highly thermodynamically favorable and, as a result, irreversible in the cell.
== History == Barbituric acid was first synthesized 27 November 1864, by German chemist Adolf von Baeyer, and later perfected in 1879 by the French chemist Edouard Grimaux, who developed and improved the synthesis. This was done by condensing urea with diethyl malonate. There are several stories about how the substance got its name. The most likely story is that Baeyer and his colleagues went to celebrate their discovery in a tavern where the town's artillery garrison were also celebrating the feast of Saint Barbara – the patron saint of artillerymen. An artillery officer is said to have christened the new substance by amalgamating Barbara with urea. Another story holds that Baeyer synthesized the substance from the collected urine of a Munich waitress named Barbara. No substance of medical value was discovered, however, until 1902 when two German scientists working at Bayer, Emil Fischer and Joseph von Mering, discovered that barbital was very effective in putting dogs to sleep. Barbital was then marketed by Bayer under the trade name Veronal. It is said that Mering proposed this name because the most peaceful place he knew was the Italian city of Verona. In 1912, Bayer introduced another barbituric acid derivative, phenobarbital, under the trade name Luminal, as a sedative–hypnotic. It was not until the 1950s that the behavioral disturbances and physical dependence potential of barbiturates became recognized. Since the 1970s, most barbiturates were replaced by benzodiazepines.
Brazil's international relations are based on Article 4 of the Federal Constitution, which establishes non-intervention, self-determination, international cooperation and the peaceful settlement of conflicts as the guiding principles of Brazil's relationship with other countries and multilateral organizations. According to the constitution, the president has ultimate authority over foreign policy, while Congress is tasked with reviewing and considering all diplomatic nominations and international treaties, as well as legislation relating to Brazilian foreign policy. Brazil's foreign policy is a by-product of the country's position as a regional power in Latin America, a leader among developing countries, and an emerging world power. Brazilian foreign policy has generally been based on the principles of multilateralism, peaceful dispute settlement, and non-intervention in the affairs of other countries. Brazil is a founding member state of the Community of Portuguese Language Countries (CPLP), also known as the Lusophone Commonwealth, an international organization and political association of Lusophone nations. An increasingly well-developed tool of Brazil's foreign policy is providing aid as a donor to other developing countries. Brazil does not just use its growing economic strength to provide financial aid, but it also provides high levels of expertise and most importantly of all, a quiet non-confrontational diplomacy to improve governance levels. Total aid is estimated to be around $1 billion per year.
Obesity in children is treated with dietary changes and physical activity, often through an intensive counselling and lifestyle changes program. At least sixty minutes of daily moderate to high intensity aerobic physical activity is recommended for all children (regardless of BMI). Dieting, including caloric restriction or very low calorie diets, and missing meals should however be discouraged, as it is associated with psychological harm, risks of dietary or nutritional deficiencies and a risk of developing eating disorders later in life. The benefit of tracking BMI and providing counselling around weight is minimal. Brief primary care weight management interventions (e.g. delivered by a physician or nurse practitioner) have only a marginal positive effect in reducing childhood overweight or obesity.

Reported activity is highest at MC4R, with lower potency at MC1R and MC3R. The MC4R interaction is generally treated as the most relevant to its central effects. Selectivity is not absolute, and activity across the family is dose-dependent.
Indirect evidence from animal studies supports central access, and the proposed mechanism requires it. Direct quantification in humans is limited. How much reaches specific brain regions remains an open question.
Published descriptions give a plasma half-life of roughly two to three hours after subcutaneous administration. Values vary with assay method and study population. The figure is an average rather than a fixed molecular property.
The code designates bremelanotide, a cyclic heptapeptide melanocortin receptor agonist. It served as an internal research identifier before the compound entered formal clinical development. The code and the generic name refer to the same molecule.
