Outside the approved product, bremelanotide circulates as a research chemical sold by peptide vendors, often labelled PT-141. Such material is not manufactured under pharmaceutical quality standards, and independent testing has repeatedly found content that differs from the label. Analytical certificates supplied with a purchase are not strong evidence of purity because they are usually generated by the seller. Online discussion tends to blur the distinction between the approved drug and unregulated powder, which complicates interpretation of reported experiences.
PT-141 is the original development code for bremelanotide, a synthetic peptide first studied as a potential tanning and sexual-response agent in the 1990s. Researchers at a small American biotechnology firm designed it as a shortened analogue of melanotan II, which itself came from work on alpha-melanocyte-stimulating hormone. Early screening focused on pigmentation, but behavioural observations in animal models redirected attention toward sexual motivation. That shift made PT-141 one of the first melanocortin compounds investigated specifically for effects on desire rather than on skin colour.
Early work on melanocortin analogs in the 1980s and 1990s produced peptides intended to influence pigmentation and appetite. One of these, melanotan II, was observed to affect sexual desire as an incidental finding in self-administration reports. Researchers then pursued analogs with altered receptor selectivity and improved handling characteristics, and PT-141 emerged from that program in the late 1990s. The development path moved from dermatology and metabolism toward a central nervous system application, a shift that shaped both trial designs and the eventual label.
Regulatory review of bremelanotide concluded in 2019 with approval in the United States for a defined indication in premenopausal women. The reviewed formulation is a single-use prefilled autoinjector given subcutaneously, and its label carries cardiovascular monitoring language tied to blood pressure changes recorded during trials. Availability outside the approving jurisdiction varies, and in several countries the compound remains unapproved or is handled as a prescription-only item. Compounded and research-grade material also circulates, and it differs from the reviewed product in purity, characterization, and chain of custody.

Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.
PT-141 is the research code for bremelanotide, a cyclic heptapeptide derived from alpha-melanocyte-stimulating hormone. The molecule belongs to the melanocortin receptor agonist family and shows highest affinity for the MC4 receptor subtype, with weaker activity at MC1, MC3 and MC5. Its structure retains the core His-Phe-Arg-Trp sequence that defines melanocortin recognition, while cyclization and terminal modifications improve metabolic stability relative to the parent hormone. Early work classified the compound as a centrally acting agent rather than a peripherally acting vasodilator, which shaped subsequent development priorities.
Identity and purity are established with orthogonal methods rather than a single test. Reverse-phase high-performance liquid chromatography with ultraviolet detection gives a purity figure by area normalization, while mass spectrometry confirms the expected molecular ion. Amino acid analysis or peptide mapping can detect sequence errors that a mass value alone would miss, and residual counterion content is sometimes measured separately. Common impurities include truncated sequences, oxidized residues, and deamidated products; reporting them individually is more informative than a single composite purity number.
Lyophilized peptide arrives as a white to off-white cake or powder and is normally held at minus twenty degrees Celsius or colder for extended periods. Short-term bench work at ambient temperature is tolerable for minutes, not hours, because the solid is hygroscopic and picks up moisture that promotes hydrolysis. Vials should stay in a desiccator or a sealed bag with desiccant, protected from light, since aromatic residues in the sequence are susceptible to photo-oxidation. Inventory records that note arrival date and storage location reduce the chance of using degraded material.
The lyophilised solid is relatively stable when kept dry, protected from light and held at reduced temperature, commonly minus 20 degrees Celsius or lower for long-term storage. In solution the peptide is more vulnerable: tryptophan oxidation, hydrolysis of the lactam bridge and aggregation all become relevant over time, and the rate depends on pH, buffer composition and concentration. Repeated freeze-thaw cycles are generally avoided because they promote aggregation. Aqueous working solutions are typically prepared fresh or split into single-use aliquots to limit degradation before analysis.
Published studies differ in design, population and endpoint definition, so results are not always directly comparable across reports. Some trials used patient-reported measures of desire and distress, while others tracked physiological or behavioural outcomes. Questions that remain open include the durability of effects beyond short follow-up periods, the frequency of transient blood pressure elevation observed after administration, and whether a subtype-selective analogue could separate central effects from pigmentation-related activity. These points are usually framed as unresolved rather than settled in review literature.
Routine characterisation of bremelanotide relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nm, where the peptide backbone absorbs. Mass spectrometry, typically in tandem mode with electrospray ionisation, confirms identity and supports quantification in biological matrices. Additional checks include amino acid analysis, peptide mapping after enzymatic digestion, and confirmation of the lactam bridge, since incomplete cyclisation produces a mass-shifted by-product. Purity values above 95 percent are common in reference-grade material, though reports vary in how strictly related substances are resolved from the parent peak.
Melanocortin receptors form a family of five G-protein-coupled proteins, labelled MC1R through MC5R, that respond to peptides derived from pro-opiomelanocortin. Alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone are the best-known endogenous ligands. The receptors are distributed differently across tissues, so a single agonist can produce effects in the brain, skin, adrenal gland and vasculature. This distribution explains why one peptide can influence both pigmentation and motivated behaviour.
Bremelanotide is a cyclic heptapeptide that binds several melanocortin receptors rather than one. In vitro assays report agonist activity at MC1R, MC3R, MC4R and MC5R, with MC4R generally treated as the subtype most relevant to sexual effects. MC4R is expressed in hypothalamic nuclei involved in appetite, energy balance and motivated behaviour, which provides a plausible route for central action. Selective MC4R agonists studied in animals produce comparable behavioural changes, supporting that interpretation.
== Inhibitors == Inhibitors of ALS are used as herbicides that slowly starve affected plants of these amino acids, which eventually leads to inhibition of DNA synthesis. They affect grasses and dicots alike. They are not a chemistry class but rather a mechanism of action class with diverse chemistries. The ALS inhibitor family includes sulfonylureas (SUs), imidazolinones, triazolopyrimidines (see Category:Triazolopyrimidines), pyrimidinyl oxybenzoates, and sulfonylamino carbonyl triazolinones. As of March 2022, the ALS inhibitors suffer the worst (known) resistance problem of all herbicide classes, having 169 known resistant target species. The structures of ALS herbicides are radically different from the normal substrate and so none of them bind at the catalytic site but instead at a site specific to herbicidal action. Therefore resistance mutations are expected to have widely varying effects on normal ALS catalysis activity, positive, negative, and neutral. Unsurprisingly that is exactly what experiments have shown, including Yu et al., 2007 finding resistance in Hordeum murinum due to a proline→serine substitution at amino acid 197 to increase ALS activity by 2x-3x.
The city and the metropolitan area has a dense transport network. This solves the problem of a break between the lower town and the upper town and the two parts of the city are connected by long boulevards, winding roads, many stairs, a funicular and the Jenner tunnel. The CODAH transport network is called Lia and is operated by the Ocean Port Transport company (CTPO), a subsidiary of Veolia Transport. The overhaul of the bus network in 2008 helped to ensure a better service for all the towns in the metropolitan area. The CTPO operates a bus network consisting of 19 regular urban routes and six evening routes called the "Midnight Bus". The Le Havre urban area is served by 165 vehicles and 41 regular bus routes with an average of 100,000 passengers per day. From January 2011 there has been a regular shuttle service specific to the Industrial Zone and Port of Le Havre, thus adding to the cross-estuary service of VTNI. Since 1890 the funicular has provided a link between the upper town and the lower town in four minutes with a cable car. Le Havre had a tramway system from 1894 until it closed in 1957. More recently a new tramway system, with 23 stations and 13 km (8 mi) of route, was built, and opened on 12 December 2012. The first part of the line connects the beach to the station climbing to the upper town through a new tunnel near the Jenner tunnel then it splits into two: one link going to Mont-Gaillard, the other to Caucriauville.
=== Glycolysis === When more R5P is needed than NADPH, R5P can be formed through glycolytic intermediates. Glucose 6-phosphate is converted to fructose 6-phosphate (F6P) and glyceraldehyde 3-phosphate (G3P) during glycolysis. Transketolase and transaldolase convert two molecules of F6P and one molecule of G3P to three molecules of R5P. During rapid cell growth, higher quantities of R5P and NADPH are needed for nucleotide and fatty acid synthesis, respectively. Glycolytic intermediates can be diverted toward the non-oxidative phase of PPP by the expression of the gene for pyruvate kinase isozyme, PKM. PKM creates a bottleneck in the glycolytic pathway, allowing intermediates to be utilized by the PPP to synthesize NADPH and R5P. This process is further enabled by triosephosphate isomerase inhibition by phosphoenolpyruvate, the PKM substrate.
=== Spectrophotometric === In spectrophotometric assays, you follow the course of the reaction by measuring a change in how much light the assay solution absorbs. If this light is in the visible region you can actually see a change in the color of the assay, and these are called colorimetric assays. The MTT assay, a redox assay using a tetrazolium dye as substrate is an example of a colorimetric assay. UV light is often used, since the common coenzymes NADH and NADPH absorb UV light in their reduced forms, but do not in their oxidized forms. An oxidoreductase using NADH as a substrate could therefore be assayed by following the decrease in UV absorbance at a wavelength of 340 nm as it consumes the coenzyme. Direct versus coupled assays

While A, T, C, and G represent a particular nucleotide at a position, there are also letters that represent ambiguity which are used when more than one kind of nucleotide could occur at that position. The rules of the International Union of Pure and Applied Chemistry (IUPAC) are as follows: For example, W means that either an adenine or a thymine could occur in that position without impairing the sequence's functionality.
=== Actual consequences === In retrospect, it is now known that smoke from the Kuwait oil fires only affected the weather pattern throughout the Persian Gulf and surrounding region during the periods that the fires were burning in 1991, with lower atmospheric winds blowing the smoke along the eastern half of the Arabian Peninsula, and cities such as Dhahran and Riyadh, and countries such as Bahrain experienced days with smoke filled skies and carbon soot rainout/fallout. Thus the immediate consequence of the arson sabotage was a dramatic regional decrease in air quality, causing respiratory problems for many Kuwaitis and those in neighboring countries. According to the 1992 study from Peter Hobbs and Lawrence Radke, daily emissions of sulfur dioxide (which can generate acid rain) from the Kuwaiti oil fires were 57% of that from electric utilities in the United States, the emissions of carbon dioxide were 2% of global emissions and emissions of soot reached 3400 metric tons per day. In a paper in the DTIC archive, published in 2000, it states that "Calculations based on smoke from Kuwaiti oil fires in May and June 1991 indicate that combustion efficiency was about 96% in producing carbon dioxide. While, with respect to the incomplete combustion fraction, Smoke particulate matter accounted for 2% of the fuel burned, of which 0.4% was soot." (With the remaining 2% being oil that did not undergo any initial combustion).
Prognosis is related to disease severity. AS can range from mild to progressively debilitating and from medically controlled to refractory. Some cases may have times of active inflammation followed by times of remission resulting in minimal disability while others never have times of remission and have acute inflammation and pain, leading to significant disability. As the disease progresses, it can cause the vertebrae and the lumbosacral joint to ossify, resulting in the fusion of the spine. This places the spine in a vulnerable state because it becomes one bone, which causes it to lose its range of motion as well as putting it at risk for spinal fractures. This not only limits mobility but reduces the affected person's quality of life. Complete fusion of the spine can lead to a reduced range of motion and increased pain, as well as total joint destruction which could necessitate a joint replacement. Osteoporosis is common in ankylosing spondylitis, both from chronic systemic inflammation and decreased mobility resulting from AS. Over a long-term period, osteopenia or osteoporosis of the AP spine may occur, causing eventual compression fractures and a back "hump". Hyperkyphosis from ankylosing spondylitis can also lead to impairment in mobility and balance, as well as impaired peripheral vision, which increases the risk of falls which can cause fracture of already-fragile vertebrae. Typical signs of progressed AS are the visible formation of syndesmophytes on X-rays and abnormal bone outgrowths similar to osteophytes affecting the spine.
The potency of cultivated specimens can vary widely per each flush (harvest). In a classic paper published by Jeremy Bigwood and M.W. Beug, it was shown that with each flush, psilocybin levels varied somewhat unpredictably but were much the same on the first flush as they were on the last flush; however, psilocin was typically absent in the first two flushes but peaked by the fourth flush, making it the most potent. Two strains were also analyzed to determine potency in caps and stems: In one strain the caps contained generally twice as much psilocybin as the stems, but the small amount of psilocin present was entirely in the stems. In the other strain, a trace of psilocin was present in the cap but not in the stem; the cap and stem contained equal amounts of psilocybin. The study concluded that the levels of psilocybin and psilocin vary by over a factor of four in cultures of P. cubensis grown under controlled conditions.
Anorexia nervosa is an eating disorder characterized by attempts to lose weight by way of starvation. A person with anorexia nervosa may exhibit a number of signs and symptoms, the type and severity of which may vary and be present but not readily apparent. Although anorexia is often recognized by the physical signs, it is a mental disorder that can occur at any body weight: in the case where BMI remains at a healthy amount in adults, atypical anorexia nervosa is diagnosed. Anorexia nervosa, and the associated malnutrition that results from the self-imposed starvation, causes complications in every major organ system in the body. It can cause permanent changes in the brain due to a lack of essential nutrients in the body. Hypokalemia, a drop in the level of potassium in the blood, is a sign of anorexia nervosa. A significant drop in potassium can cause abnormal heart rhythms, constipation, fatigue, muscle damage, and paralysis. Signs and symptoms may be classified in various categories including: physical, cognitive, affective, behavioral and perceptual:

PT-141 is a research code for bremelanotide, a cyclic peptide that activates melanocortin receptors. It was developed for sexual dysfunction and later approved under a brand name as a subcutaneous injection. The same code is widely used by suppliers selling non-pharmaceutical material.
Both are synthetic melanocortin agonists and share a similar peptide backbone, but they are distinct molecules with different receptor profiles. Melanotan II was never approved as a medicine, whereas bremelanotide was. Findings about one compound should not be transferred to the other without direct evidence.
It is typically distributed as a lyophilized powder in sealed vials, sometimes with a separate solvent. Purity statements usually originate from the supplier rather than an independent laboratory. Storage recommendations vary between vendors, which makes comparison across sources difficult.
PT-141 was the internal development code assigned to bremelanotide during preclinical research. Reference sources sometimes index the peptide under the code rather than the generic name. The two terms describe the same molecule.
