This is a working overview of melanocortin receptor, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-01-01 and is reviewed periodically as new material appears.
The melanocortin system comprises five G protein-coupled receptors, designated MC1 through MC5, that signal mainly through cyclic AMP accumulation. MC1R and MC2R are associated with pigmentation and adrenal steroid production, while MC3R and MC4R are expressed in the central nervous system and influence energy balance and behavior. MC5R appears in exocrine tissues. Natural agonists include alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone, and endogenous antagonists such as agouti-related protein modulate the same sites. This receptor family provides the framework within which bremelanotide activity is described.
Bremelanotide acts as an agonist at several melanocortin receptors, with the strongest reported activity at MC4R and measurable activity at MC1R and MC3R. Because MC4R is expressed in hypothalamic and limbic circuits, the proposed mechanism links receptor activation to modulation of central pathways involved in desire rather than to direct effects on peripheral genital tissue. The precise downstream steps remain incompletely characterized, and evidence for the involvement of specific neurotransmitters is suggestive rather than settled. Nausea and blood pressure elevation reported during trials are consistent with melanocortin signaling outside the intended target circuit.
Compared with melanotan II, bremelanotide is a smaller cyclic peptide with a more constrained backbone, which affects receptor selectivity and metabolic stability. Published descriptions give a plasma half-life on the order of a few hours after subcutaneous administration, with elimination through hepatic and renal routes and limited plasma protein binding. Central access is inferred from effects observed in animal models, although direct measurement in humans is limited. Handling and storage requirements follow from the peptide backbone, which is susceptible to hydrolysis and oxidation.
Early work on melanocortin analogs in the 1980s and 1990s produced peptides intended to influence pigmentation and appetite. One of these, melanotan II, was observed to affect sexual desire as an incidental finding in self-administration reports. Researchers then pursued analogs with altered receptor selectivity and improved handling characteristics, and PT-141 emerged from that program in the late 1990s. The development path moved from dermatology and metabolism toward a central nervous system application, a shift that shaped both trial designs and the eventual label.
Regulatory review of bremelanotide concluded in 2019 with approval in the United States for a defined indication in premenopausal women. The reviewed formulation is a single-use prefilled autoinjector given subcutaneously, and its label carries cardiovascular monitoring language tied to blood pressure changes recorded during trials. Availability outside the approving jurisdiction varies, and in several countries the compound remains unapproved or is handled as a prescription-only item. Compounded and research-grade material also circulates, and it differs from the reviewed product in purity, characterization, and chain of custody.
Bremelanotide is a synthetic cyclic heptapeptide developed under the research code PT-141. The code reflects its position in an internal compound series rather than a chemical classification, and the name bremelanotide was later adopted for regulatory filings. Structurally it belongs to the melanocortin peptide family and shares a core sequence motif with alpha-melanocyte-stimulating hormone. The compound is supplied as an acetate salt in aqueous solution for injection. In reference literature it is indexed under both the code and the generic name, a dual listing that can complicate database searches.

Clinical research typically uses randomised, double-blind, placebo-controlled designs. The most common primary endpoint is the desire domain score of the Female Sexual Function Index, sometimes paired with a distress measure. Secondary outcomes include arousal, satisfaction, and event-based counts of satisfying sexual episodes. Across trials, average improvements are modest and individual responses vary widely. Whether benefits persist beyond a few months, and whether they depend on baseline hormone status, remain open questions rather than settled findings.
Melanocortin receptors form a family of five G-protein-coupled receptors designated MC1 through MC5. Bremelanotide binds most strongly at MC4R and MC1R, with weaker activity reported at MC3R and MC5R. MC4R is expressed in hypothalamic nuclei that coordinate energy balance and aspects of sexual behaviour. The prevailing interpretation is that central MC4R activation, rather than peripheral vascular effects alone, drives the reported changes in desire. This account remains partly inferential, since direct receptor-level measurement in living humans is not practical.
After subcutaneous administration, plasma concentrations rise within roughly thirty minutes and the elimination half-life is short, on the order of two to three hours. Reported physiological responses include transient increases in blood pressure and nausea, which tended to diminish with repeated dosing in trial settings. Because the peptide clears quickly, effects are not expected to persist long after a dose. Absorption from non-injected routes is poorly characterised, and nasal delivery produced variable plasma levels in older work.
Approved use is narrow and jurisdiction-specific. In the United States, the injectable product is authorised for premenopausal women with acquired, generalised hypoactive sexual desire disorder, a diagnosis that requires documented distress. It is not approved for men, for postmenopausal women, or for use alongside hormonal contraceptives under the approved labelling. Outside regulated markets, the same peptide is frequently sold as a research chemical, where identity, purity, and sterility are not independently verified.
Bremelanotide is a synthetic cyclic heptapeptide developed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and appetite signalling. Its structure contains seven amino acid residues joined by a lactam bridge that closes the ring between two side chains. The molecular formula is C50H68N14O10 and the nominal molecular mass is near 1025 daltons. Much of the early laboratory literature refers to the same molecule by the development code PT-141.
Aspartic acid was first discovered in 1827 by Auguste-Arthur Plisson and Étienne-Ossian Henry by hydrolysis of asparagine, which had been isolated from asparagus juice in 1806. Their original method used lead hydroxide, but various other acids or bases are now more commonly used instead. There are two forms or enantiomers of aspartic acid. The name "aspartic acid" can refer to either enantiomer or a mixture of two. Of these two forms, only one, "L-aspartic acid", is directly incorporated into proteins. The biological roles of its counterpart, "D-aspartic acid" are more limited. Where enzymatic synthesis will produce one or the other, most chemical syntheses will produce both forms, "DL-aspartic acid", known as a racemic mixture. In the human body, aspartate is most frequently synthesized through the transamination of oxaloacetate. The biosynthesis of aspartate is facilitated by an aminotransferase enzyme: the transfer of an amine group from another molecule such as alanine or glutamine yields aspartate and an alpha-keto acid.
=== Legal status in weapons === In 1996, the International Court of Justice (ICJ) gave an advisory opinion on the "legality of the threat or use of nuclear weapons". This made it clear, in paragraphs 54–56, that international law on poisonous weapons—the Second Hague Declaration of 29 July 1899, Hague Convention IV of 18 October 1907 and the Geneva Protocol of 17 June 1925—did not cover nuclear weapons, because their prime or exclusive use was not to poison or asphyxiate. This ICJ opinion was about nuclear weapons, but the sentence "The terms have been understood, in the practice of States, in their ordinary sense as covering weapons whose prime, or even exclusive, effect is to poison or asphyxiate," also removes depleted uranium weaponry from coverage by the same treaties as their primary use is not to poison or asphyxiate, but to destroy materiel and kill soldiers through kinetic energy. The Sub-Commission on Prevention of Discrimination and Protection of Minorities of the United Nations Human Rights Commission, passed two motions—the first in 1996 and the second in 1997. They listed weapons of mass destruction, or weapons with indiscriminate effect, or of a nature to cause superfluous injury or unnecessary suffering and urged all states to curb the production and the spread of such weapons. Included in the list was weaponry containing depleted uranium. The committee authorized a working paper, in the context of human rights and humanitarian norms, of the weapons. The requested UN working paper was delivered in 2002 by Y. K. J.
This is a list of prices of chemical elements. Listed here are mainly average market prices for bulk trade of commodities. Data on elements' abundance in Earth's crust is added for comparison. As of 2025, the most expensive non-synthetic element by mass is rhodium, and by volume, iridium. It is followed by rhodium, caesium, iridium and palladium by mass and iridium, gold and platinum by volume. Carbon in the form of diamond can be more expensive than rhodium. Per-kilogram prices of some synthetic radioisotopes range to trillions of dollars. While the difficulty of obtaining macroscopic samples of synthetic elements in part explains their high value, there has been interest in converting base metals to gold (chrysopoeia) since ancient times, but only deeper understanding of nuclear physics has allowed the actual production of a tiny amount of gold from other elements for research purposes as demonstrated by Glenn Seaborg. However, both this and other routes of synthesis of precious metals via nuclear reactions is orders of magnitude removed from economic viability. Chlorine, sulfur and carbon (as coal) are cheapest by mass. Hydrogen, nitrogen, oxygen and chlorine are cheapest by volume at atmospheric pressure. When there is no public data on the element in its pure form, price of a compound is used, per mass of element contained. This implicitly puts the value of compounds' other constituents, and the cost of extraction of the element, at zero. For elements whose radiological properties are important, individual isotopes and isomers are listed.

Among the known sons of Naram-Sin were his successor Shar-Kali-Sharri, Nabi-Ulmaš, who was governor of Tutub, and a Ukin-Ulmash. Excavations at Tell Mozan (ancient Urkesh) brought to light a sealing of Tar'am-Agade, a previously unknown daughter of Naram-Sin, who was possibly married to an unidentified endan (ruler) of Urkesh. A recently found cylinder seal, looted from Urasagrig, shows that the governor there, Sharatigubishin, was also a son. Other known children include Enmenana the "zirru priestess of the god Nanna, spouse of the god N[anna], entu priestess of the god Sin at Ur", Šumšani ēntum-priestess of Shamash at Sippar, a son who was governor at Marad, an unnamed daughter who was ēntum-priestesses at Nippur, Bin-kali-šarrē, Lipit-ilē (governor at Marad), Rigmuš-ālsu, Me-Ulmaš, and Ukēn-Ulmaš and a granddaughter Lipus-ia-um who was known to have been a lyre player for the god Sin. One daughter, Tuṭṭanabšum (Tudanapšum), held the position of high priestess of Enlil at Nippur, the most important religious position in the empire. She was also deified, the only female and only non-king to be made a god.
Outside of the main cities, signposted walks follow rivers and the escarpment of the Pennines, which is scaled in meandering stages and tunnels by the recreational Leeds-Liverpool Canal and Rochdale Canal, navigable by barge, canoe or kayak. The Yorkshire Sculpture Park and The Hepworth in Wakefield are major national art attractions, while Haworth is visited for being the home of the Bronte Sisters. Ilkley, Otley, Hebden Bridge and Holmfirth are popular smaller towns featuring farmer's markets, breweries, tea rooms, stately homes and are popular with hikers.
=== Herbivores === Herbivores consume ionone-containing carotenoids and convert those to retinal. Some species, including cattle and horses, have measurable amounts of β-carotene circulating in the blood, and stored in body fat, creating yellow fat cells. Most species have white fat and no β-carotene in circulation.
==== Gene knockout studies ==== The large number of mouse compared to human FPR receptors makes it difficult to extrapolate human FPR1 functions based on genetic (e.g. gene knockout or forced overexpression) or other experimental manipulations of FPR receptors in mice. In any event, targeted disruption of the Fpr1 gene reduced the ability of mice to survive intravenous injection of the bacterial pathogen, listeria monocytogenes; disruption of the Fpr2 gene in mice produce a similar effect while disruption of both genes further lowered the survival of mice to the listeria challenge. The effect of these gene knockouts appeared due to faulty leukocyte function and other causes leading to a breakdown in the innate immune response. The functions of the human FPR1 receptor may be equivalent to the overlapping functions of the mouse Fpr1 and Fpr2 functions and therefore be critical in the defense against at least certain bacteria. Targeted disruption of FPR-rs1 produced a 33% reduction in the lifetime of mice; there was no specific pathology associated with this reduction.

Mahathir's political views have shifted during his lengthy career. During the 1980s and 1990s, he was a supporter of third-worldism, while during other periods he has been a proponent of "Asian values" and globalisation. A Muslim thinker, he holds Islamic political views. In 2002, he characterised himself as an Islamic fundamentalist. Mahathir is generally respected in developing and Islamic countries, particularly due to his oversight of Malaysia's economic growth and his support of liberal Muslim values. Mahathir has been described as a proponent of Malay nationalism. In The Malay Dilemma, he argued that the Malay race had been marginalised, and voiced his support for affirmative action policies for them. Upon his first resignation, he expressed his disappointment at the progress made towards his "principle task" of supporting the Malay race. In 2021, Mahathir said he did not believe in Ketuanan Melayu, calling it a "fantasy", and said instead that he believed in the concept of "Bangsa Malaysia". He has been described as anti-royalist by the left-wing French newspaper Libération, owing to his efforts to oppose immunity for members of Malaysia's monarchies. Mahathir is a vocal critic of neoliberalism and the Western world. In 2011, Mahathir suggested that the September 11 attacks might have been staged by the United States government. Mahathir condemned the Universal Declaration of Human Rights in 1997, suggesting it be revised to place greater importance on economic growth over civil liberties.
== Matching gunshot residue to a specific source == If the ammunition used was specifically tagged in some way by special elements, it is possible to know the cartridge used to produce the gunshot residue. Inference about the source of gunshot residue can be based on the examination of the particles found on a suspect and the population of particles found on the victim, in the firearm or in the cartridge case, as suggested by the ASTM Standard Guide for gunshot residue analysis by scanning electron microscopy/energy dispersive X-ray spectrometry. Advanced analytical techniques such as ion beam analysis (IBA), carried out after scanning electron microscopy, can support further information allowing one to infer about the source of gunshot residue particles. Christopher et al. showed as the grouping behaviour of different makes of ammunition can be determined using multivariate analysis. Bullets can be matched back to a gun using comparative ballistics.
At times of immobility, muscle wasting in bears is also suppressed by a proteolytic inhibitor that is released in circulation. Another factor that contributes to the sustaining of muscle strength in hibernating bears is the occurrence of periodic voluntary contractions and involuntary contractions from shivering during torpor. The three to four daily episodes of muscle activity are responsible for the maintenance of muscle strength and responsiveness in bears during hibernation.
==== Part-time work ==== Approximately 30% of women in France work part-time, compared to 5% of men. Of the 4.1 million part-time workers, 83% are women. The prevalence of part-time work among women is closely associated with the presence of young children in the household. Among part-time workers, 28% of women and 42% of men report working part-time involuntarily, indicating a preference for more working hours.
Clinical attachment level (CAL) is a clinical measurement used in periodontology to determine the position of the periodontal attachment relative to a fixed anatomical landmark on the tooth, usually the cementoenamel junction (CEJ). It is a fundamental parameter for assessing the severity and progression of periodontal disease, monitoring treatment outcomes, and evaluating changes in periodontal support over time. Clinical attachment loss refers to the pathological loss of periodontal attachment, which is quantified by the clinical attachment level. Unlike probing depth alone, clinical attachment level accounts for changes in the position of the gingival margin, providing a more accurate assessment of periodontal attachment loss.

Reported activity is highest at MC4R, with lower potency at MC1R and MC3R. The MC4R interaction is generally treated as the most relevant to its central effects. Selectivity is not absolute, and activity across the family is dose-dependent.
Indirect evidence from animal studies supports central access, and the proposed mechanism requires it. Direct quantification in humans is limited. How much reaches specific brain regions remains an open question.
Published descriptions give a plasma half-life of roughly two to three hours after subcutaneous administration. Values vary with assay method and study population. The figure is an average rather than a fixed molecular property.
PT-141 was the internal development code assigned to bremelanotide during preclinical research. Reference sources sometimes index the peptide under the code rather than the generic name. The two terms describe the same molecule.
