PT-141 is the original development code for bremelanotide, a synthetic peptide first studied as a potential tanning and sexual-response agent in the 1990s. Researchers at a small American biotechnology firm designed it as a shortened analogue of melanotan II, which itself came from work on alpha-melanocyte-stimulating hormone. Early screening focused on pigmentation, but behavioural observations in animal models redirected attention toward sexual motivation. That shift made PT-141 one of the first melanocortin compounds investigated specifically for effects on desire rather than on skin colour.
Clinical development proceeded through two routes of administration. An intranasal formulation advanced first, but variable absorption and tolerability problems led to a switch to subcutaneous injection. The United States Food and Drug Administration approved the subcutaneous product in 2019 for hypoactive sexual desire disorder in premenopausal women. Marketing rights subsequently changed hands, and commercial availability has fluctuated since approval. Use in men, in postmenopausal women, and in combination with other agents remains outside the approved label.
Outside the approved product, bremelanotide circulates as a research chemical sold by peptide vendors, often labelled PT-141. Such material is not manufactured under pharmaceutical quality standards, and independent testing has repeatedly found content that differs from the label. Analytical certificates supplied with a purchase are not strong evidence of purity because they are usually generated by the seller. Online discussion tends to blur the distinction between the approved drug and unregulated powder, which complicates interpretation of reported experiences.
The peptide contains seven amino acids arranged in a ring, closed by a lactam bridge between a side-chain acid and an amine. This cyclic constraint holds the backbone in a defined conformation and increases resistance to enzymatic breakdown relative to linear analogs. N-terminal acetylation and a C-terminal amide further protect the molecule from exopeptidases. The result is a compound with a comparatively long circulation time for a small peptide. Structural modification of the bridge alters receptor affinity, which is one reason analogs in this family differ in their subtype preferences.
Whether the behavioral effect originates centrally, peripherally, or through both remains an active question. Animal experiments using receptor antagonists and site-specific injections point toward hypothalamic melanocortin circuits as a key locus, but translating those findings to humans is not straightforward. Blood pressure changes observed in trials suggest a vascular component that may be peripherally mediated. The relationship between receptor occupancy and reported effect has not been mapped in humans, and no validated biomarker predicts response. This gap makes it difficult to explain individual variability on pharmacological grounds alone.
Bremelanotide acts as an agonist at melanocortin receptors, a family of five G-protein-coupled receptors labeled MC1 through MC5. Binding studies indicate activity at several of these subtypes rather than strict selectivity for one. Signalling proceeds mainly through Gs-mediated activation of adenylyl cyclase, raising intracellular cyclic AMP. The MC4 receptor, expressed in hypothalamic and limbic circuits, is widely regarded as the subtype most relevant to sexual response. Because the molecule is not subtype-selective, effects at other melanocortin receptors are expected and are used to explain some observed side effects.

The compound binds several melanocortin receptor subtypes rather than a single target, with the strongest functional activity reported at MC4R and measurable activity at MC1R, MC3R and MC5R. MC4R populations are dense in hypothalamic nuclei that integrate energy balance, autonomic tone and reproductive behaviour, which is the anatomical basis for the proposed pro-desire effect. Because binding is not subtype-selective, pigmentary and vascular effects accompany central activity. Improving subtype selectivity is an active area of analogue design. Direct causal mapping from receptor occupancy to reported desire change in humans is not fully established.
After subcutaneous dosing, peak plasma concentrations appear within roughly one hour, and elimination is fast, with a half-life on the order of a few hours. Degradation is mainly proteolytic, and at least one circulating fragment retains receptor activity, so parent-drug levels alone do not describe total exposure. Clearance does not depend heavily on hepatic cytochrome enzymes, which lowers the likelihood of common metabolic interaction routes. Data in renal or hepatic impairment are limited. Repeated dosing does not appear to produce marked accumulation given the short half-life.
Reported pharmacodynamic effects include transient rises in blood pressure and heart rate, flushing, nausea and headache, appearing soon after dosing and resolving within hours. These responses were dose-related in early studies and shaped the label's cardiovascular cautions and blood pressure monitoring advice. Gastrointestinal upset is the most frequent reason cited for discontinuation in trials. Whether the vascular signal attenuates with repeated use is not settled. Central effects on desire are described as emerging over weeks rather than immediately, which points to a cumulative rather than acute process.
Bremelanotide acts as an agonist at several melanocortin receptors, with the strongest reported activity at MC4R and measurable activity at MC1R and MC3R. Because MC4R is expressed in hypothalamic and limbic circuits, the proposed mechanism links receptor activation to modulation of central pathways involved in desire rather than to direct effects on peripheral genital tissue. The precise downstream steps remain incompletely characterized, and evidence for the involvement of specific neurotransmitters is suggestive rather than settled. Nausea and blood pressure elevation reported during trials are consistent with melanocortin signaling outside the intended target circuit.
Compared with melanotan II, bremelanotide is a smaller cyclic peptide with a more constrained backbone, which affects receptor selectivity and metabolic stability. Published descriptions give a plasma half-life on the order of a few hours after subcutaneous administration, with elimination through hepatic and renal routes and limited plasma protein binding. Central access is inferred from effects observed in animal models, although direct measurement in humans is limited. Handling and storage requirements follow from the peptide backbone, which is susceptible to hydrolysis and oxidation.
The melanocortin system comprises five G protein-coupled receptors, designated MC1 through MC5, that signal mainly through cyclic AMP accumulation. MC1R and MC2R are associated with pigmentation and adrenal steroid production, while MC3R and MC4R are expressed in the central nervous system and influence energy balance and behavior. MC5R appears in exocrine tissues. Natural agonists include alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone, and endogenous antagonists such as agouti-related protein modulate the same sites. This receptor family provides the framework within which bremelanotide activity is described.
== Timeline of detections == NPI was first identified in the UK in August 2024 by WEDINOS as samples W054816 and W057074, then the full chemical characterisation and first literature reported sample was provided by CanTEST in Australia in September 2024,. NPI was later detected in the Netherlands in May 2025, and in Finland in early 2025.
== Cellular immunotherapy == Immune cell-based therapies for solid tumors may be limited by tumor-antigen heterogeneity, restricted immune-cell trafficking, an immunosuppressive tumor microenvironment, limited cell persistence, treatment-related toxicities, and complex manufacturing requirements.
The practice of reading a sentient's mind without permission (something the Culture is technologically easily capable of) is strictly taboo. The whole plot of Look to Windward relies on a Hub Mind not reading an agent's mind (with certain precautions in case this rule gets violated). Minds that do so anyway are considered deviant and shunned by other Minds (see GCU Grey Area). At one point it is said that if the Culture actually had written laws, the sanctity of one's own thoughts against the intrusion of others would be the first on the books. This gives some measure of privacy and protection; though the very nature of Culture society would, strictly speaking, make keeping secrets irrelevant: most of them would be considered neither shameful nor criminal. It does allow the Minds in particular to scheme amongst themselves in a very efficient manner, and occasionally withhold information.
While the main habitat of earthworms is in soil, they are not restricted to this habitat. The brandling worm Eisenia fetida lives in decaying plant matter and manure. Arctiostrotus vancouverensis from Vancouver Island and the Olympic Peninsula is generally found in decaying conifer logs. Aporrectodea limicola, Sparganophilus spp., and several others are found in mud in streams (riparian species). Some species are arboreal, some aquatic and some euryhaline (salt-water tolerant) and littoral (living on the sea-shore), e.g. Pontodrilus litoralis. Even in the soil species, special habitats, such as ultramafic soils derived from serpentine, have an earthworm fauna of their own, e.g. New Caledonian Acanthodrilidae. Vermicomposting of organic wastes is currently processed using the epigeic Eisenia fetida and Eisenia andrei which are particularly adapted to the consumption of fresh organic matter without deepening in the substrate (contrary to the anecis Lumbricus terrestris) and which tolerate the heat generated in compost heaps. Addition of this humified organic matter to the soil, preferably as a surface mulch, will improve moisture and nutrient availability, with beneficial effects on the soil microflora. It will also provide several species of earthworms with their food and nutrient requirements, and will create the optimum conditions of temperature and moisture that will stimulate their activity, although to a lesser extent than bacterial-mediated compost. Earthworms are environmental indicators of soil health.

=== Alexandria Ship Project === In January 2016, a mid-18th century ship was discovered on the waterfront of Alexandria, Virginia. Conservation efforts include keeping the wood submerged and wet since the wooden frame was waterlogged. The conservation of this ship is ongoing. When the waterlogged wooden frame was originally removed from the archaeological site, the timber frames were stored in fresh water vats until June 2017. The framed structure was then packaged and sent to the Conservation Research Laboratory at Texas A&M University for conservation. Documentation and conservation included laser scanning, modeling, X-ray, and wood degradation analysis before the treatment using polyethylene glycol and vacuum freeze drying
=== Urinary tract infection === People with diabetes show an increased rate of urinary tract infection. The reason is that bladder dysfunction is more common in people with diabetes than in people without diabetes due to diabetes nephropathy. When present, nephropathy can cause a decrease in bladder sensation, which in turn can cause increased residual urine, a risk factor for urinary tract infections.
The major surviving part of the walls is visible adjacent to the only remaining gateway under the tower of the Church of St John the Baptist. By the mid-14th century Bristol is considered to have been England's third-largest town (after London and York), with an estimated 15–20,000 inhabitants on the eve of the Black Death of 1348–49. The plague inflicted a prolonged demographic setback, with the population estimated at between 10,000 and 12,000 during the 15th and 16th centuries. One of the first great merchants of Bristol was William Canynge. Born c. 1399, he was five times mayor of the town and twice represented it as an MP. He is said to have owned ten ships and employed over 800 sailors. In later life he became a priest and spent a considerable part of his fortune in rebuilding St Mary Redcliffe church, which had been severely damaged by lightning in 1446.

This abuse of power by local leaders exacerbated the violent purges and terror campaigns carried out by Stalin against members of the party deemed to be traitors. With the Great Purge (1936–1938), Stalin rid himself of internal enemies in the party and rid the Soviet Union of any alleged socially dangerous and counterrevolutionary person who might have offered legitimate political opposition to Marxism–Leninism. Stalin allowed the secret police NKVD (People's Commissariat for Internal Affairs) to rise above the law and the GPU (State Political Directorate) to use political violence to eliminate any person who might be a threat, whether real, potential, or imagined. As an administrator, Stalin governed the Soviet Union by controlling the formulation of national policy, but he delegated implementation to subordinate functionaries. Such freedom of action allowed local communist functionaries much discretion to interpret the intent of orders from Moscow, but this allowed their corruption. To Stalin, the correction of such abuses of authority and economic corruption were responsibility of the NKVD. In the 1937–1938 period, the NKVD arrested 1.5 million people, purged from every stratum of Soviet society and every rank and file of the party, of which 681,692 people were killed as enemies of the state.

== Adverse effects == The most common effects when taking dapoxetine are nausea, dizziness, dry mouth, headache, diarrhea, and insomnia. Discontinuation rates due to adverse effects and costs are very high, as demonstrated in a study in Asia which showed that cumulative discontinuation rates within one year are as high as 87%. Unlike other SSRIs used to treat depression, which have been associated with high incidences of sexual dysfunction, dapoxetine is associated with low rates of sexual dysfunction. Taken as needed, dapoxetine has very mild adverse effects of decreased libido (<1%) and erectile dysfunction (<4%).
== Domains == The SH2 domain of Grb2 binds to phosphorylated tyrosine-containing peptides on receptors or scaffold proteins with a preference for pY-X-N-X, where X is generally a hydrophobic residue such as valine (see [1]). The N-terminal SH3 domain binds to proline-rich peptides and can bind to the Ras-guanine exchange factor SOS. The C-terminal SH3 domain binds to peptides conforming to a P-X-I/L/V/-D/N-R-X-X-K-P motif that allows it to specifically bind to proteins such as Gab-1.

PT-141 is a research code for bremelanotide, a cyclic peptide that activates melanocortin receptors. It was developed for sexual dysfunction and later approved under a brand name as a subcutaneous injection. The same code is widely used by suppliers selling non-pharmaceutical material.
Both are synthetic melanocortin agonists and share a similar peptide backbone, but they are distinct molecules with different receptor profiles. Melanotan II was never approved as a medicine, whereas bremelanotide was. Findings about one compound should not be transferred to the other without direct evidence.
It is typically distributed as a lyophilized powder in sealed vials, sometimes with a separate solvent. Purity statements usually originate from the supplier rather than an independent laboratory. Storage recommendations vary between vendors, which makes comparison across sources difficult.
It is frequently described as an MC4 receptor agonist, but binding assays show activity at more than one melanocortin subtype. Selectivity is therefore relative rather than absolute. This matters when interpreting side effects tied to other receptor subtypes.
