Bremelanotide is a synthetic cyclic heptapeptide that acts on a family of G-protein-coupled receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide associated with pigmentation and several central signalling pathways. A lactam bridge constrains the ring and slows enzymatic breakdown, which distinguishes it from the linear parent molecule. Research interest moved over time from pigment biology toward central nervous system effects, particularly circuits connected to sexual desire. Parenteral delivery is used because oral bioavailability is poor.
Early clinical work used an intranasal formulation, and later programmes switched to subcutaneous delivery for more consistent absorption. A subcutaneous product received regulatory approval in the United States in 2019 for premenopausal women with acquired, generalised hypoactive sexual desire disorder. Approval followed phase 3 trials in which active treatment separated from placebo on desire and distress measures, though the average difference was modest. Labeling carries a caution about transient blood pressure elevation, so cardiovascular history is assessed before prescribing. Questions about durability of benefit beyond several months remain open.
Published discussion sits at the intersection of peptide chemistry, neuroendocrinology and sexual medicine. Trial reports emphasise change scores on validated instruments, while mechanistic papers focus on hypothalamic circuits and receptor selectivity. Because placebo response in this field is large, effect sizes are usually reported with confidence intervals rather than as isolated averages. Reviews note that female and male data sets are not interchangeable and should be read separately. Diagnostic terminology has been revised over time, which complicates comparison between older and newer studies.
Melanocortin receptors form a family of five G-protein-coupled receptors designated MC1 through MC5. Bremelanotide binds most strongly at MC4R and MC1R, with weaker activity reported at MC3R and MC5R. MC4R is expressed in hypothalamic nuclei that coordinate energy balance and aspects of sexual behaviour. The prevailing interpretation is that central MC4R activation, rather than peripheral vascular effects alone, drives the reported changes in desire. This account remains partly inferential, since direct receptor-level measurement in living humans is not practical.
After subcutaneous administration, plasma concentrations rise within roughly thirty minutes and the elimination half-life is short, on the order of two to three hours. Reported physiological responses include transient increases in blood pressure and nausea, which tended to diminish with repeated dosing in trial settings. Because the peptide clears quickly, effects are not expected to persist long after a dose. Absorption from non-injected routes is poorly characterised, and nasal delivery produced variable plasma levels in older work.

Clinical development proceeded through two routes of administration. An intranasal formulation advanced first, but variable absorption and tolerability problems led to a switch to subcutaneous injection. The United States Food and Drug Administration approved the subcutaneous product in 2019 for hypoactive sexual desire disorder in premenopausal women. Marketing rights subsequently changed hands, and commercial availability has fluctuated since approval. Use in men, in postmenopausal women, and in combination with other agents remains outside the approved label.
Outside the approved product, bremelanotide circulates as a research chemical sold by peptide vendors, often labelled PT-141. Such material is not manufactured under pharmaceutical quality standards, and independent testing has repeatedly found content that differs from the label. Analytical certificates supplied with a purchase are not strong evidence of purity because they are usually generated by the seller. Online discussion tends to blur the distinction between the approved drug and unregulated powder, which complicates interpretation of reported experiences.
Published studies differ in design, population and endpoint definition, so results are not always directly comparable across reports. Some trials used patient-reported measures of desire and distress, while others tracked physiological or behavioural outcomes. Questions that remain open include the durability of effects beyond short follow-up periods, the frequency of transient blood pressure elevation observed after administration, and whether a subtype-selective analogue could separate central effects from pigmentation-related activity. These points are usually framed as unresolved rather than settled in review literature.
Routine characterisation of bremelanotide relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nm, where the peptide backbone absorbs. Mass spectrometry, typically in tandem mode with electrospray ionisation, confirms identity and supports quantification in biological matrices. Additional checks include amino acid analysis, peptide mapping after enzymatic digestion, and confirmation of the lactam bridge, since incomplete cyclisation produces a mass-shifted by-product. Purity values above 95 percent are common in reference-grade material, though reports vary in how strictly related substances are resolved from the parent peak.
The lyophilised solid is relatively stable when kept dry, protected from light and held at reduced temperature, commonly minus 20 degrees Celsius or lower for long-term storage. In solution the peptide is more vulnerable: tryptophan oxidation, hydrolysis of the lactam bridge and aggregation all become relevant over time, and the rate depends on pH, buffer composition and concentration. Repeated freeze-thaw cycles are generally avoided because they promote aggregation. Aqueous working solutions are typically prepared fresh or split into single-use aliquots to limit degradation before analysis.
Handling procedures emphasize minimizing moisture uptake, since the dried solid is hygroscopic and can draw water when warmed to room temperature before a vial is opened. Laboratories record lot number, purity, counter-ion content, and residual solvent data supplied by the producer. Impurity profiles are compared run to run, and any shift in retention time or peak shape prompts re-verification against a reference standard. Certificates of analysis are treated as claims to be checked rather than accepted at face value.
Identity and purity testing for this peptide typically relies on reversed-phase high-performance liquid chromatography with ultraviolet detection, reported as area-percent purity. Mass spectrometry, usually in tandem mode, confirms molecular mass and supports quantification in biological matrices. Sequence confirmation may use peptide mapping after enzymatic digestion, while nuclear magnetic resonance and circular dichroism supply supplementary structural detail. No single technique establishes identity alone, so laboratories compare retention time, mass, and fragment pattern against a verified reference standard.
Valve is an American video game developer and publisher founded in 1996 by Gabe Newell and Mike Harrington. The company is based in Bellevue, Washington. Valve's first game was Half-Life, a first-person shooter released in 1998. It sold over nine million retail copies. Alongside Half-Life's launch, Valve released development tools to enable the player community to create content and mods. The company then proceeded to hire the creators of popular mods such as Counter-Strike. Valve continued their trend of developing predominantly first-person video games in the 2000s with a number of critically successful releases. In 2004, they released the highly anticipated sequel Half-Life 2 through their own digital distribution service Steam. The game sold over 10 million copies and was met with acclaim. Valve released two subsequent episodes for Half-Life 2 and later packaged those games together with the puzzle game Portal and the multiplayer shooter Team Fortress 2 in a collection known as The Orange Box. By the end of 2008, combined retail sales of the Half-Life series, Counter-Strike series and The Orange Box had surpassed 32 million units. Newell also projected that digital sales of Valve's games would eventually exceed retail sales as Steam continued to grow. In the late 2000s, Valve released two zombie-themed first-person shooters focusing on cooperative gameplay with the Left 4 Dead series. The company continued to release multiplayer games with the launches of Counter-Strike: Global Offensive and Dota 2, both of which have large esports communities fostered by Valve.
=== Mycolic acid analysis === Mycolic acid analysis has been an evolving field of study for gas-liquid chromatography, as it offers a solution to slow growth rates in Mycobacterium. Mycolic acid is a fatty acid found in the disease tuberculosis, offering a chemical target for diagnosticians to look for.
=== Foreign policy shift === From the outset, the Aznar government was committed to greater Spanish involvement in international actions. Thus, the need to seek a new model of Armed Forces that would make them more operational was raised, which, together with the spectacular growth of conscientious objector inclined the PP towards the formula of an exclusively professional army by putting an end to compulsory military service ─ thus abandoning the mixed model implemented by the Socialists.

Disulfiram inhibits the enzyme acetaldehyde dehydrogenase, which in turn results in buildup of acetaldehyde, a toxic metabolite of ethanol with unpleasant effects. The medication or drug is commonly used to treat alcohol use disorder, and results in immediate hangover-like symptoms upon consumption of alcohol, this effect is widely known as disulfiram effect.
Peukert stated "the conquest of the world by a secularized, scientific rationality was so overwhelming, that the switch from religion to science as the main source of a meaning-creating mythology for everyday life took place almost without resistance. The result, however, was that science took upon itself a burden of responsibility that it would soon find a heavy one". Peukert wrote science could not offer spiritual consolation as in a world dominated by science the question of "how can the rationalist, secular ideal of the greatest happiness of the greatest number be vindicated, given that it is rebutted in the case of each individual by illness, suffering and death?", which was impossible to answer. As such, scientists came to be concerned with the body of the individual as a way of determining if that individual should be allowed to pass on his/her genes to the next generation with the criterion being whatever the individual was of "value" or not. In this way, there was a shift from the individual as the center of medical concern to the collective of the volkskörper (the "body" of the entire race).
=== Bleeding and blood clot risk === Cirrhosis can increase the risk of bleeding. The liver produces various proteins in the coagulation cascade (coagulation factors II, VII, IX, X, V, and VI). When damaged, the liver is impaired in its production of these proteins. This will ultimately increase bleeding as clotting factors are diminished. Clotting function is estimated by lab values, mainly platelet count, prothrombin time (PT), and international normalized ratio (INR). The American Gastroenterological Association (AGA) provided recommendations in 2021 in regards to coagulopathy management of cirrhotic patients in certain scenarios.
=== Food === Fumaric acid has been used as a food acidulant since 1946. It is approved for use as a food additive in the EU, USA, Australia, and New Zealand. As a food additive, it is used as an acidity regulator and can be denoted by the E number E297. It is generally used in beverages and baking powders for which requirements are placed on purity. Fumaric acid is used in the making of wheat tortillas as a food preservative and as the acid in leavening. It is generally used as a substitute for tartaric acid and occasionally in place of citric acid, at a rate of 1 g of fumaric acid to every ~1.5 g of citric acid, in order to add sourness, similarly to the way malic acid is used. As well as being a component of some artificial vinegar flavors, such as "Salt and Vinegar" flavored potato chips, it is also used as a coagulant in stove-top pudding mixes. The European Commission Scientific Committee on Animal Nutrition, part of DG Health, found in 2014 that fumaric acid is "practically non-toxic" but high doses are likely nephrotoxic after long-term use.

=== Livestock and crop predation === A limitation of food sources in early spring and wild berry and nut crop failures in summer may contribute to bears regularly feeding from human-based food sources. These bears often eat crops, especially during autumn hyperphagia when natural foods are scarce. Favored crops include apples, oats and corn. American black bears can do extensive damage in areas of the northwestern United States by stripping the bark from trees and feeding on the cambium. Livestock depredations occur mostly in spring. Although they occasionally hunt adult cattle and horses, they seem to prefer smaller prey such as sheep, goats, pigs and young calves. They usually kill by biting the neck and shoulders, though they may break the neck or back of the prey with blows with the paws. Evidence of a bear attack includes claw marks and is often found on the neck, back and shoulders of larger animals. Surplus killing of sheep and goats is common. American black bears have been known to frighten livestock herds over cliffs, causing injuries and death to many animals; whether this is intentional is not known. Occasionally bears kill pets, especially domestic dogs, which are most prone to harass a bear. It is not recommended to use unleashed dogs to deter bear attacks. Although large, aggressive dogs can sometimes cause a bear to run, if pressed, angry bears often turn the tables and end up chasing the dogs in return. A bear in pursuit of a pet dog can threaten both canid and human lives.
Cyanocobalamin is a form of vitamin B12 used to treat and prevent vitamin B12 deficiency except in the presence of cyanide toxicity. The deficiency may occur in pernicious anemia, following surgical removal of the stomach, with fish tapeworm, or due to bowel cancer. It is given by mouth, by injection into a muscle, or as a nasal spray. Cyanocobalamin is generally well tolerated. Minor side effects may include diarrhea, nausea, upset stomach, and itchiness. Serious side effects may include anaphylaxis, and low blood potassium resulting in heart failure. Use is not recommended in those who are allergic to cobalt or have Leber's disease. No overdosage or toxicity has been recorded by the FDA. It is less preferred than hydroxocobalamin for treating vitamin B12 deficiency because it has a slightly lower bioavailability. Some studies have shown it to possess an antihypotensive effect. Vitamin B12 is an essential nutrient meaning that it cannot be made by the body but is required for life. Cyanocobalamin was first manufactured in the 1940s. It is available as a generic medication and over the counter. In 2023, it was the 104th most commonly prescribed medication in the United States, with more than 6 million prescriptions.
=== Launch === The chain was announced in July 2023. The first CosMc's store opened on December 7, 2023 in the village of Bolingbrook, Illinois. The opening day saw a large queue of customers in the drive-thru which was the only way to order through the restaurant. CNN reported that the Bolingbrook location saw more than twice the number of visits for a typical McDonald's location, citing data from Placer.ai, with the customer base largely skewed to those aged 22–29. At the time of launch, McDonald's planned to have ten operational stores, including nine in Texas by 2024. McDonald's CEO Chris Kempczinski said that the company would evaluate the ten stores as a "limited test”, adding "Let's not get too excited." In its first month, the store reportedly had twice the traffic of a regular McDonald's. A new app and rewards program, CosMc’s Club, along with mobile ordering, was introduced in May 2024. In early 2025, the company announced it would close three of its larger format locations and open two smaller locations in Texas.
During World War II the Germans experimented with the idea of the schnorchel (snorkel) from captured Dutch submarines but did not see the need for them until rather late in the war. The schnorchel is a retractable pipe that supplies air to the diesel engines while submerged at periscope depth, allowing the boat to cruise and recharge its batteries while maintaining a degree of stealth. Especially as first implemented however, it turned out to be far from a perfect solution. There were problems with the device's valve sticking shut or closing as it dunked in rough weather. Since the system used the entire pressure hull as a buffer, the diesels would instantaneously suck huge volumes of air from the boat's compartments, and the crew often suffered painful ear injuries. Speed was limited to 8 knots (15 km/h), lest the device snap from stress. The schnorchel also created noise that made the boat easier to detect with sonar, yet more difficult for the on-board sonar to detect signals from other vessels. Finally, allied radar eventually became sufficiently advanced that the schnorchel mast could be detected beyond visual range. In clear weather, diesel exhausts can be seen on the surface to a distance of about three miles, while "periscope feather" (the wave created by the snorkel or periscope moving through the water) is visible from far off in calm sea conditions. Modern radar is also capable of detecting a snorkel in calm sea conditions.
Andexanet alfa is a biologic agent, a recombinant modified version of human activated factor X (FXa). Andexanet alfa differs from native FXa due to the removal of a 34 residue fragment that contains the Gla domain. This modification reduces andexanet alfa's anticoagulant potential. Additionally, a serine to alanine (S419A) mutation in the active site eliminates its activity as a prothrombin to thrombin catalyst, but still allows the molecule to bind to FXa inhibitors. FXa inhibitors bind to andexanet alfa with the same affinity as to natural FXa. As a consequence, in the presence of andexanet alfa, natural FXa is partially freed, which can lead to effective hemostasis. In other words, it acts as a decoy receptor. Andexanet alfa reverses effect of all anticoagulants that act directly through FXa or by binding antithrombin III. The drug is not effective against factor IIa inhibitor dabigatran. Its activity is measured using the anti-Xa test, which is utilized to determine the amount of available factor Xa for coagulation

It is a synthetic cyclic peptide that activates melanocortin receptors. It is given by injection and was approved in the United States in 2019 for a specific low-desire diagnosis in premenopausal women. It is not a hormonal therapy.
Earlier approaches were largely hormonal or psychological in focus, whereas this compound acts on central melanocortin signalling. It is not a vasodilator and does not share the mechanism of phosphodiesterase inhibitors. Head-to-head comparative data are limited.
Placebo responses in desire trials are large, so the average drug-placebo separation is small. Reporting therefore relies on validated questionnaires with statistical ranges. Individual responses vary widely.
Trial results generally show a small to moderate average improvement in desire scores relative to placebo. The distribution of responses is wide, and some participants show little measurable change. Group averages should not be read as a prediction for any single person.
